Scientist II, Discovery

 

Lassogen is an early-stage biotechnology company in San Diego that is passionate about establishing lasso peptides as a new class of programmable therapeutics. Lasso peptides are defined by an unusual 3D threaded structure with a knotted topology that engenders them with superior properties such as high stability and activity against a growing number of disease targets that are challenging for existing modalities such as antibodies and small molecules. We currently are advancing lasso peptides that target G protein-coupled receptors, with applications in oncology, autoimmunity, and pain and inflammation.


We are seeking a Scientist II to develop synthetic biology methods aimed at discovery and optimization of novel lasso therapeutics. Areas of responsibility will include strain engineering for lasso peptide production, cell-free biosynthesis for the discovery of lasso peptides, lasso peptide evolution to optimize properties, and protein engineering to optimize pathway enzymes. Key Responsibilities:

  • Design, build, and engineer lasso peptide biosynthetic pathways in various production hosts, including Streptomyces, Pseudomonas, Burkholderia, and C. glutamicum

  • Engineer cells using rational approaches to overproduce lasso peptides, including lead candidates for in vitro and in vivo testing.

  • Express lasso peptide biosynthetic gene clusters using cell-free biosynthesis technology for rapid discovery and evolution of lasso peptide analogs.

  • Create, characterize, and screen diverse lasso peptide libraries based on computational designs and display technologies, including the introduction of non-natural amino acids.

  • Overexpress and purify lasso peptide biosynthetic enzymes. Engineer lasso peptide biosynthetic enzymes to improve activity and stability.

  • Acquire, evaluate, interpret, and present experimental results internally, at partner meetings, and at scientific conferences.

  • Execute safe laboratory procedures and wear personal protection equipment at all times in the labs.

  • Follow and help enforce all protocols associated with maintaining safe operations in the labs and offices during the COVID-19 pandemic.


Qualifications and Skills:

  • Ph.D. (or equivalent experience) in molecular biology, synthetic biology, microbiology, cell biology, or related disciplines (non-Ph.D. scientists with extensive relevant experience also will be considered).  Postdoctoral work in these disciplines is strongly preferred.   

  • Experience designing, refactoring, expressing, and optimizing natural product biosynthetic pathways.

  • Skilled in engineering and cultivating a range of bacterial strains (e.g., E. coli, Streptomyces, Burkholderia, etc.).

  • Expert in recombinant DNA techniques and tools for genetically manipulating microorganisms of different types (e.g., DNA design, cloning, isolation, and sequencing).

  • Proficient in bioinformatic analysis and DNA visualization (e.g., Geneious, Vector NTI, etc.), BLAST search, sequence alignment, genomic database search, and related software.

  • Familiar with Next-Gen DNA sequencing (NGS) methods.

  • Experience with protein purification, protein pull-down assays, and Western analysis

  • Experience with protein evolution, display technologies (e.g., phage display, DNA display, RNA display, ribosome display, etc.), and bio-panning procedures are desirable.

  • Strong publication history in peer-reviewed journals.

  • Able to thrive in a fast-paced, dynamic, multi-disciplinary, team-driven work environment.

  • Must be detail-oriented, self-motivated, and able to work independently.

  • Excellent organization, strong interpersonal skills, excellent oral and written communication skills. Able to manage, organize, and critically evaluate data.

Work Environment:
This position is based at Lassogen’s facility within the San Diego Science Center. This position primarily will involve working in a laboratory and office setting and will require communication with other individuals from different backgrounds through a variety of methods, both remote and in-person. Travel may be required periodically.

 
Associate Scientist, Synthetic Biology

Natural lasso peptides are produced by bacteria, and Lassogen research is exploiting these biosynthetic pathways to advance lasso peptides for treating human disease.


We are seeking an Associate Scientist with synthetic biology experience to join a team focused on discovery and optimization of novel lasso therapeutics. Areas of responsibility will include strain engineering for lasso peptide production, lasso peptide screening using cell-free biosynthesis, and protein engineering to optimize pathway enzymes. Candidates will continuously interact with a multi-disciplinary team and make important contributions to our groundbreaking science in an exciting start- up environment where everyone makes a difference.   • Cultivate bacteria, yeast, and mammalian cells for lasso peptide production and assay development

  • Support the R&D team to:

    • Engineer cells to overproduce lasso peptides

    • Express lasso peptide biosynthetic gene clusters using cell-free biosynthesis technology for high throughput discovery of natural lasso peptides and evolution of lasso peptides

    • Create, characterize, and screen diverse lasso peptide libraries based on computational designs, including the introduction of non-natural amino acids

  • Acquire, evaluate, interpret, and present experimental results

  • Follow laboratory safety procedures and wear personal protection equipment at all times in the lab


Qualifications and Skills:

  • BS or MS (or equivalent experience) in molecular biology, synthetic biology, microbiology, cell biology or related disciplines

  • Experienced in recombinant DNA techniques (e.g., DNA design, cloning, isolation, and sequencing)

  • Skilled in cultivating bacteria (e.g. , )

  • Skilled in expressing genes and gene clusters is host organisms

  • Proficient in DNA visualization software (e.g., Geneious, Vector NTI, etc.) and bioinformatic tools, (e.g. BLAST, sequence alignment, genomic database search).

  • Familiarity with HPLC/LCMS methods for metabolite identification and quantification is a plus

  • Able to manage, organize, and critically evaluate data.

  • Able to thrive in a fast-paced, dynamic, multi-disciplinary, team-driven work environment.

  • Must be detail-oriented, self-motivated, and able to work independently.

  • Excellent organizational skills, strong interpersonal skills, excellent oral and written communication skills.
